Not sure with Proftpd, but with samba server, you can add additional shares in /etc/samba/smb.conf:

[drive2]
   comment = DietPi Share 2
   path = /mnt/location_to_drive2
   browseable = yes
   create mask = 0775
   directory mask = 0775
   valid users = root
   writeable = yes

Can samba and proftpd run at the same time?

Yep, you can install both through DietPi-Software. Set Fileserver choice to none/manual, Go start install.

After reboot select ProFTP and Samba server from the DietPi optimized software list ( ────File Servers at bottom). Go start install
